中药研究中在线二维液相色谱的应用(3)

Chen等[46]建立了基于CMC的全二维液相色谱分析方法,通过两个loop环分别往复收集第一维色谱中的馏分,经切换阀往复切换到第二维分析柱上进行分析,从而达到全二维分析模式。如采用人肝癌细胞HepG2 / CMC柱(流速0. 2 mL/ min,500μL loop环,每2. 5 min切换一次)和RP 18e硅胶整体柱 (流速3. 5 mL/ min)-TOF MS系统,从黄柏[46]、苦参[46]和鸦胆子[47]中筛选出具有抗肝癌活性的 成 分;第 一 维 采 用 人 慢 性 髓 原 白 血 病 细 胞K562 / CMC柱,从青黛中筛选得到活性较好的抗白血病化合物[48].通过改进,将正常心肌细胞/ CMC柱和DOX(多柔比星)诱导心衰细胞/ CMC柱联用,并依次作为第一维色谱柱,从附子中特征地筛选得到对DOX-诱导心力衰竭有效的4种附子生物碱类化合物[49].为从口服黄芩的大鼠血浆中筛选出潜在的抗肝癌活性物质,在第一维采用HepG2 / CMC柱的基础上,将loop环换为XDB-C18柱(12. 5 mm×4. 6 mm,5 μm),对口服黄芩大鼠的血浆样品进行在线预处理和富集[50],从而建立筛选中药在生物体内有效物质的CMC-全二维液相分析方法。
3结论与展望
在线二维液相色谱因峰容量高、分离效率好而受到众多研究者的青睐。近年来,在线二维液相色谱被广泛地应用于多个研究领域中复杂样品的分析。在线二维液相法在中药中的应用显着地提高了中药化学成分分析和识别的效率,为后续药效物质基础、作用机制和质量评价研究奠定了良好基础。
